There is an urgent need for rare diseases such as Amyotrophic Lateral Sclerosis (ALS). The high heterogeneity of disease courses and severe disability urges the development of surrogate markers for use in clinical trials. We use multi-contrast 7T MRI to allow an individualized brain and spinal cord mapping of regional neurodegeneration and we combine it with biomarkers of neuronal loss such as neurofilament light chain levels to model subclinical progression in a longitudinal setting.
